Ein literarischer Stoff. Layla ist die Figur aus «The Story of Layla and Majnun» vom persischen Schriftsteller Nizami Ganjavi, der im 12. Jahrhundert lebte. Eric Clapton war von diesem Stoff angetan, weil er in den Protagonisten seine eigene unmögliche Liebe zu Patti Boyd, der Ehefrau von Musiker-Freund George Harrison widerspiegelt sah.
Duane Allman spielte zweite Leadgitarre. Clapton und Allman wurden einander von Tom Dowd (Atlantic Records vorgestellt. Sie wurden Freunde und spielten zusammen, wobei sich herausstellte, dass sie so gut harmonierten, als würden sie schon sehr lange miteinander musizieren.
Strukturell ist der Song bemerkenswert, weil die Contrasting Verse-Chorus Struktur zuerst in eine Extended Coda mündet (2:18–3:07) und dann in ein anderes namenloses Stück übergeht, ein instrumentales Piano-Stück, das vom Schlagzeuger Jim Gordon komponiert wurde und das Clapton an Layla
